New signing David Miller struck fifty from just 33 balls, to help Glamorgan move to the top of the south group table.
Captain Jacques Rudolph also hit fifty as he and Miller put on 85 for the third wicket, which laid the foundations for Glamorgan's innings as they ended on 176/5.
Despite a typically belligerent innings from Klinger (52) Gloucestershire were always behind the run rate thanks to some fantastic bowling from the away side as they secured a comfortable 25 run victory.
It was a good team performance from the Welsh County, but it was man of the match David Miller who was the catalyst behind the win.
"I'm very happy to be one (win) from one, and looking forward to the next four games. I was a bit nervous for a couple of balls, it took me six or seven to get going but it's nice batting with Jacques Rudolph who has a lot of experience.”
Miller’s motto is ‘if it’s in the arc, it’s out the park’ and this proved to be the case yesterday as he smashes three sixes during his innings.
"It's always good to hit the pavilion, the one I got out I was trying to hit over the top of the apartments, but it's nice to hit a few sixes and get the run-rate up.
"Every single game I get nervous, but it's what you do with the nerves and where you take it - it's a short stint to make an impression but I'm just trying to play my game and enjoy it, which has been really easy with this changing-room."
